Background of Nestin

Nestin is a type VI intermediate filament  (IF) protein that in humans is encoded by the NES gene. This gene is mapped to 1q23.1. Distribution and expression of nestin in mitotically active cells suggests it plays a role in regulation of the assembly and disassembly of intermediate filaments, which, together with other structural proteins, participate in remodeling of the cell. The role of nestin in dynamic cells, particularly structural organization of the cell, appears strictly regulated by phosphorylation, especially its integration into heterogeneous intermediate filaments together with vimentin or α-internexin. Furthermore, nestin expression has been extensively used as a marker for central nervous system (CNS) progenitor cells in different contexts, based on observations indicating a correlation between nestin expression and this cell type in vivo.

Antibody Validation

Boster validates all antibodies on WB, IHC, ICC, Immunofluorescence, and ELISA with known positive control and negative samples to ensure specificity and high affinity, including thorough antibody incubations.

Asked: 2020-04-01

Answer

The anti-Nestin antibody (PB9874) has not been validated for cross reactivity specifically with primate tissues, though there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in primate you can get your next antibody for free. Please contact me if I can help you with anything.

Asked: 2017-12-05

Answer

Based on literature cervix carcinoma does express NES. Based on Uniprot.org, NES is expressed in metanephric glomerulus, placenta, neuroblastoma, cervix carcinoma, liver, cervix carcinoma erythroleukemia, among other tissues. Regarding which tissues have NES expression, here are a few articles citing expression in various tissues:
Cervix carcinoma, Pubmed ID: 17081983, 18669648, 20068231
Cervix carcinoma, and Erythroleukemia, Pubmed ID: 23186163
Liver, Pubmed ID: 18318008, 24275569
Neuroblastoma, Pubmed ID: 12880961

Question

We have tried in the past anti-Nestin antibody for WB on metanephric glomerulus a few years ago. I am using mouse, and We intend to use the antibody for IHC next. My question regards examining metanephric glomerulus as well as neuroblastoma in our next experiment. Could you please give me some suggestion on which antibody would work the best for IHC?

Asked: 2017-08-15

Answer

I have checked the website and datasheets of our anti-Nestin antibody and it seems that PB9874 has been validated on mouse in both WB and IHC. Thus PB9874 should work for your application. Our Boster satisfaction guarantee will cover this product for IHC in mouse even if the specific tissue type has not been validated. We do have a comprehensive range of products for IHC detection and you can check out our website bosterbio.com to find out more information about them.
